چکیده

Accompanying the autoxidation of hydroxylamine at pH 10.2, nitroblue tetrazolium was reduced and nitrite was produced in the presence of EDTA. The rate of at&oxidation was negligible below pH 8.0, but sharply increased with increasing pH. The reduction of nitroblue tetrazolium was inhibited by euperoxide dismutaee, indicating the participation of superoxide anion radical in the autoxidation. Hydrogen peroxide stimulated the autoxidation and superoxide diemutase inhibited the hydrogen peroxide-induced oxidation, results which suggest the participation of hydrogen peroxide in autoxidation and in the generation of superoxide radical. An assay for superoxide dismutase using autoxidation of hydroxylamine is described.

The role of superoxide anion in the autoxidation of epinephrine and a simple assay for superoxide dismutase.

The rate of autoxidation of epinephrine and the sensitivity of this autoxidation to inhibition by superoxide dismutase were both augmented, as the pH was raised from 7.8 --f 10.2. 02-, generated by the xanthine oxidase reaction, caused the oxidation of epinephrine to adrenochrome and the yield of adrenochrome produced per 02introduced, increased with increasing pH in the range 7.8 + 10.2 and al...

Kinetic investigations of the autoxidation of adrenalin.

Generation rates of superoxide anions (O2-) by autoxidizing adrenalin at pH 9.5 were determined in solutions containing either superoxide dismutase (0.85 M-1 S-1) or hydroxylamine (0.0185 M-1 S-1) as competitive scavengers. The rate constants of O2- with adrenalin and hydroxylamine were calculated for neutral and alkaline solutions. The respective values were for adrenalin: 5.6 X 10(4) M-1 S-1,...

The generation of hydrogen peroxide, superoxide radical, and hydroxyl radical by 6-hydroxydopamine, dialuric acid, and related cytotoxic agents.

Hydrogen peroxide, superoxide radical, and hydroxyl radical were all formed during the autoxidation of four cytotoxic agents, namely, 6-hydroxydopamine, 6-aminodopamine, 6,7-dihydroxytryptamine, and dialuric acid. Ascorbic acid and 5-hydroxydopamine, two autoxidizable agents of biological interest but without similar cytotoxic actions, were used as negative controls. The results tend to implica...

Enhancement of mitochondrial, cyanide-resistant superoxide dismutase in the livers of rats treated with 2,4-dinitrophenol.

Homogenates prepared from the livers of rats treated for 3 days with the uncoupling agent 2,4-dinitrophenol contained considerably higher cyanide-sensitive superoxide dismutase activity when compared with homogenates of saline-treated animals. The effect was specific for only one form of the enzyme, however. Administration of 2,4-dinitrophenol at a dosage and on a schedule which produced the cl...
